Bovine Corticotropin Releasing Factor ELISA Kit
Principle of the Assay: CRF ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CRF antibody and an CRF-HRP conjugate. The assay sample and buffer are incubated together with CRF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CRF concentration since CRF from samples and CRF-HRP conjugate compete for the anti-CRF antibody binding site. Since the number of sites is limited, as more sites are occupied by CRF from the sample, fewer sites are left to bind CRF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CRF concentration in each sample is interpolated from this standard curve.

Corticotropin-releasing hormone is secreted by the paraventricular nucleus (PVN) of the hypothalamus in response to stress. Marked reduction in this protein has been observed in association with Alzheimer disease and autosomal recessive hypothalamic corticotropin deficiency has multiple and potentially fatal metabolic consequences including hypoglycemia and hepatitis. In addition to production in the hypothalamus, this protein is also synthesized in peripheral tissues, such as T lymphocytes and is highly expressed in the placenta. In the placenta it is a marker that determines the length of gestation and the timing of parturition and delivery. A rapid increase in circulating levels of the hormone occurs at the onset of parturition, suggesting that, in addition to its metabolic functions, this protein may act as a trigger for parturition.
