Molecular Structure
A soluble fusion protein consisting of the extracellular (134aa) domain of human CD28 fused to murine IgG2a Fc (233aa).

Performance
CD28-muIg was tested for activity and quantitated in EIA. Fusion protein was captured using Goat-anti-mouse antibody coated plate. After 45 minute incubation, plates were washed twice and an excess of Mouse Ig was added to completely block the plate. Anti-CD28/Biotin followed by Streptavidin/HRP, and TMB/H2 O2 were used for detection. Fusion protein was detectable at 4 ng/ml in this assay.
Production
Fusion protein was Protein A purified from (low FBS containing) tissue culture supernatant of CHO transfectants. Purity was >95% by SDS-PAGE with less than 1% Bovine Immunoglobulin. Product was 0.2 um filtered and vialed under aseptic conditions.
Buffer
50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl. Product was sterile filtered and vialed under aseptic conditions.
Preparation and Storage
Store at 2 - 5 degree C. Open under aseptic conditions. Freeze/Thawing is not recommended.
Product should retain activity for at least 12 months after shipping date when stored as recommended.

Information: Human CD28 is an important costimulatory molecule found on all CD4+ T cells and on about half of the CD8+T cells. T cell activities attributed to CD28 include, prevention of anergy, induction of cytokine gene transcription, stabilization of cytokine mRNAs and activation of CD8+ cytotoxic T lymphocytes. The ligands for CD28, identified as CD80 (B7-1) and CD86 (B7-2), are immunoglobulin superfamily monomeric transmembrane glycoproteins of 60 kd and 80 kd, respectively.
