Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cyclin-dependent kinase 7. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Cyclin-dependent kinase 7 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Cyclin-dependent kinase 7, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Cyclin-dependent kinase 7 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

CDK7: a protein kinase of the CDK family. Forms a trimeric complex with cyclin H and MAT1, which functions as a Cdk-activating kinase (CAK). Activates the cyclin-associated kinases CDK1, -2, -4 and -6. An essential component of the transcription factor TFIIH, that is involved in transcription initiation and DNA repair. Serves as a direct link between the regulation of transcription and the cell cycle. Phosphorylates and activates RNA polymerase II, allowing its escape from the promoter and elongation of the transcripts. Involved in cell cycle control and in RNA transcription by RNA polymerase II. Its expression and activity are constant throughout the cell cycle.
