Intended Uses: This sandwich kit is for the accurate quantitative detection of human Bcl2/Adenovirus E1B 19kDa Interacting Protein 3 (also known as BNIP3) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human BNIP3 antibody. BNIP3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human BNIP3 Antibody is added and binds to BNIP3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated BNIP3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human BNIP3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Involved in mitochondrial quality control via its interaction with SPATA18/MIEAP: in response to mitochondrial damage, participates in mitochondrial protein catabolic process (also named MALM) leading to the degradation of damaged proteins inside mitochondria. The physical interaction of SPATA18/MIEAP, BNIP3 and BNIP3L/NIX at the mitochondrial outer membrane regulates the opening of a pore in the mitochondrial double membrane in order to mediate the translocation of lysosomal proteins from the cytoplasm to the mitochondrial matrix. Plays an important role in the calprotectin (S100A8/A9)-induced cell death pathway.
