Principle of the Assay: The Rat E Cadherin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat E Cadherin in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Rat E Cadherin coated on a 96-well plate. Standards and samples are pipetted into the wells and E Cadherin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat E Cadherin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of E Cadherin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Cadherin-1, also known as CAM 120/80 or epithelial cadherin (E-cadherin) or uvomorulin, is a protein that in humans is encoded by the CDH1 gene. E-cadherin is a Ca(2+)-dependent epithelial cell-cell adhesion molecule. Downregulation of E-cadherin expression often correlates with strong invasive potential and poor prognosis of human carcinomas. The gene spans a region of approximately 100 kb, and its location on chromosome 16q22.1. It contains 16 exons and a 65-kb-long intron 2. E-cadherin gene mutations may contribute to the development of diffusely growing gastric carcinomas. It plays a central part in the process of epithelial morphogenesis and acts as a strong invasion suppressor in experimental tumor cell systems. The standard product used in this kit is recombinant gene expression with the molecular mass of 120KDa.

CDH1: a single-pass type I membrane protein, and calcium dependent cell adhesion proteins. It is a ligand for integrin alpha-E/beta-7, and it colocalizes with DLG7 at sites of cell-cell contact in intestinal epithelial cells. Anchored to actin microfilaments through association with alpha-, beta- and gamma-catenin. Sequential proteolysis induced by apoptosis or calcium influx, results in translocation from sites of cell-cell contact to the cytoplasm. Involved in mechanisms regulating cell-cell adhesions, mobility and proliferation of epithelial cells. Defects in CDH1 are involved in dysfunction of the cell-cell adhesion system, triggering cancer invasion (gastric, breast, ovary, endometrium and thyroid) and metastasis. Has a potent invasive suppressor role.
