Specificity
This assay has high sensitivity and excellent specificity for detection of CAV-1. No significant cross-reactivity or interference between CAV-1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CAV-1 and all the analogues, therefore, cross reaction may still exist in some cases.

Assay Type
Quantitative Sandwich

Intended Uses: This CAV-1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CAV-1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CAV-1 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for CAV-1. Standards or samples are then added to the microtiter plate wells and CAV-1 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of CAV-1 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for CAV-1 are added to each well to "sandwich" the CAV-1 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain CAV-1 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CAV-1 concentration in each sample is interpolated from this standard curve.

Uniprot Description

Function: Involved in the costimulatory signal essential for T-cell receptor (TCR)-mediated T-cell activation. Its binding to DPP4 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner. May act as a scaffolding protein within caveolar membranes. Interacts directly with G-protein alpha subunits and can functionally regulate their activity. Forms a stable heterooligomeric complex with CAV2 that targets to lipid rafts and drives caveolae formation. Recruits CTNNB1 to caveolar membranes and may regulate CTNNB1-mediated signaling through the Wnt pathway. Ref.7

Subunit structure: Homooligomer

By similarity. Interacts (via the N-terminus) with DPP4; the interaction is direct

By similarity. Forms a stable heterooligomeric complex with CAV2 that targets to lipid rafts and drives caveolae formation. Interacts with BMX, BTK, CTNNB1, CDH1, GLIPR2, JUP, NOSTRIN, SNAP25 and STX1A

By similarity. Interacts with PACSIN2. Ref.11

Subcellular location: Golgi apparatus membrane; Peripheral membrane protein

By similarity. Cell membrane; Peripheral membrane protein

By similarity. Membrane › caveola; Peripheral membrane protein. Membrane raft

By similarity. Note: Colocalized with DPP4 in membrane rafts. Potential hairpin-like structure in the membrane. Membrane protein of caveolae

By similarity. Ref.11

Post-translational modification: The N-terminus of both isoforms are blocked.Phosphorylated at Tyr-14 by ABL1 in response to oxidative stress. Ref.8 Ref.9

Sequence similarities: Belongs to the caveolin family.

Product Notes

The Mouse Cav-1 cav1 (Catalog #AAA721447) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA721447 ELISA Kit recognizes Mouse Cav-1. It is sometimes possible for the material contained within the vial of "Caveolin-1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
