Canine Inter-alpha-trypsin inhibitor heavy chain H2 (ITIH2) ELISA Kit
Principle of the Assay||ITIH2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ITIH2 antibody and an ITIH2-HRP conjugate. The assay sample and buffer are incubated together with ITIH2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ITIH2 concentration since ITIH2 from samples and ITIH2-HRP conjugate compete for the anti-ITIH2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ITIH2 from the sample, fewer sites are left to bind ITIH2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ITIH2 concentration in each sample is interpolated from this standard curve.

The inter-alpha-trypsin inhibitors (ITI) are a family of structurally related plasma serine protease inhibitors involved in extracellular matrix stabilization and in prevention of tumor metastasis. The ITI family contains multiple proteins made up of a light chain (see MIM 176870) and a variable number of heavy chains (Salier et al., 1987 [PubMed 2446322]; Himmelfarb et al., 2004 [PubMed 14744536]).[supplied by OMIM, Nov 2009]
