Principle of the Assay: CA-12 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CA-12 antibody and an CA-12-HRP conjugate. The assay sample and buffer are incubated together with CA-12-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CA-12 concentration since CA-12 from samples and CA-12-HRP conjugate compete for the anti-CA-12 antibody binding site. Since the number of sites is limited, as more sites are occupied by CA-12 from the sample, fewer sites are left to bind CA-12-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CA-12 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is one of several isozymes of carbonic anhydrase, which catalyzes reversible hydration of carbon dioxide. Defects in this enzyme are associated with osteopetrosis and renal tubular acidosis. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jun 2014]

Competitively inhibited by saccharin, thioxolone, coumarins, 667-coumate, celecoxib (Celebrex), valdecoxib (Bextra), SC-125, SC-560, diclofenac, acetate, azide, bromide, sulfonamide derivatives such as acetazolamide (AZA), methazolamide (MZA), ethoxzolamide (EZA), dichlorophenamide (DCP), brinzolamide, dansylamide, thiabendazole-5-sulfonamide, trifluoromethane sulfonamide and N-hydroxysulfamide, fructose-based sugar sulfamate RWJ-37497, and Foscarnet (phosphonoformate trisodium salt). Repressed strongly by hydrogen sulfide(HS) and weakly by nitrate (NO(3)). Esterase activity weakly reduced by cyanamide. N- hydroxyurea interfers with zinc binding and inhibit activity. Belongs to the alpha-carbonic anhydrase family.
