INTENDED USE This BMP-6 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine BMP-6. This ELISA kit for research use only
PRINCIPLE OF THE ASSAY BMP-6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BMP-6 antibody and an BMP-6-HRP conjugate. The assay sample and buffer are incubated together with BMP-6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BMP-6 concentration since BMP-6 from samples and BMP-6-HRP conjugate compete for the anti-BMP-6 antibody binding site. Since the number of sites is limited, as more sites are occupied by BMP-6 from the sample, fewer sites are left to bind BMP-6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BMP-6 concentration in each sample is interpolated from this standard curve.

The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development. In addition, the fact that this BMP is closely related to BMP5 and BMP7 has lead to speculation of possible bone inductive activity. [provided by RefSeq, Jul 2008]
