Related Product Information for p40-phox blocking peptide
Nicotinamide adenine dinucleotide phosphate (NADPH)-oxidase is a multimeric enzyme system that mediates electron transport from NADPH in the cytoplasm to molecular oxygen in the phagosome, thereby generating reactive oxidant intermediates. Upon neutrophil stimulation, NADPH-oxidase and other cytosolic elements localize to the cell membrane from the cytosol to form a complex which produces phagocytic oxygen radicals. There are a number of cytosolic proteins that are involved in NADPH-oxidase activation/deactivation, including p47-phox, p67-phox, p40-phox and the small GTP-binding protein, Rac. Activation of NADPH oxidase is accompanied by the phosphorylation of cytosolic components p40-phox, p47-phox and p67-phox. The PKC consensus phosphorylation sites Thr 154 and Ser 315 in p40-phox are phosphorylated during activation of NADPH oxidase. p40-phox can promote oxidase activation by increasing the affinity of p47-phox for NADPH-oxidase.

NCBI Description

The protein encoded by this gene is a cytosolic regulatory component of the superoxide-producing phagocyte NADPH-oxidase, a multicomponent enzyme system important for host defense. This protein is preferentially expressed in cells of myeloid lineage. It interacts primarily with neutrophil cytosolic factor 2 (NCF2/p67-phox) to form a complex with neutrophil cytosolic factor 1 (NCF1/p47-phox), which further interacts with the small G protein RAC1 and translocates to the membrane upon cell stimulation. This complex then activates flavocytochrome b, the membrane-integrated catalytic core of the enzyme system. The PX domain of this protein can bind phospholipid products of the PI(3) kinase, which suggests its role in PI(3) kinase-mediated signaling events. The phosphorylation of this protein was found to negatively regulate the enzyme activity. Alternatively spliced transcript variants encoding distinct isoforms have been observed. Uniprot Description

p40phox: a component of the NADPH-oxidase, a multicomponent enzyme system responsible for the oxidative burst. Upon neutrophil stimulation, this protein and other cytosolic elements are sent to the cell membrane from the cytosol to form a complex which produces phagocytic oxygen radicals. Responsible for the downregulation of NADPH-oxidase. Alternative splicing has been observed.

Product Notes

The p40-phox ncf4 (Catalog #AAA3001306) is a Blocking Peptide and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's p40-phox can be used in a range of immunoassay formats including, but not limited to, Blocking. Researchers should empirically determine the suitability of the p40-phox ncf4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "p40-phox, Blocking Peptide" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
