Intended Uses: This AT2R ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine AT2R. This ELISA kit for research use only!

Principle of the Assay: AT2R ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AT2R antibody and an AT2R-HRP conjugate. The assay sample and buffer are incubated together with AT2R-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AT2R concentration since AT2R from samples and AT2R-HRP conjugate compete for the anti-AT2R antibody binding site. Since the number of sites is limited, as more sites are occupied by AT2R from the sample, fewer sites are left to bind AT2R-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AT2R concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the G-protein coupled receptor 1 family, and functions as a receptor for angiotensin II. It is an intergral membrane protein that is highly expressed in fetus, but scantily in adult tissues, except brain, adrenal medulla, and atretic ovary. This receptor has been shown to mediate programmed cell death and this apoptotic function may play an important role in developmental biology and pathophysiology. Mutations in this gene are been associated with X-linked mental retardation. [provided by RefSeq, Jan 2010]
