Caspase 6 Assay Kit (Colorimetric) is based on the hydrolysis of the peptide substrate Ac-VEID-pNA (acetyl-Val-Glu-Ilel-Asp p-nitroanilide) by caspase 6, resulting in the release of the p-nitroaniline (pNA), which can be quantified using a spectrophotometer or a microtiter plate reader at at 405 nm.
Preparation and Storage
Stable for at least 12 months at recommended temperature from date of shipment.

Description: Caspase-6 Assay Kit (Colorimetric) provides a simple and convenient method for detecting the activity of caspase-6.

Background: Caspase family of cysteine proteases has been shown to play a key role in apoptosis. Mammalian caspases can be subdivided into three functional groups: initiator caspases (Caspase 2, 8, 9 and 10), executioner caspases (Caspase-3, 6 and 7), and inflammatory caspases (Caspase 1, 4, 5, 11 and 12). Caspase-6 (also known as Mch2) is a member of the caspase-family of cysteine proteases. During apoptosis procaspase-6 is processed at aspartate residues by self-proteolysis and/or cleavage by another caspase. The processed active form of caspase-6 consists of large (18 kDa) and small (11 kDa) subunits which associate to form the active enzyme. Active caspase-6 has been shown involving in the proteolysis of PARP (poly ADP-ribose polymerase), an enzyme that is involved in DNA repair and genomic maintenance.
