Principle of the assay: AR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AR antibody and an AR-HRP conjugate. The assay sample and buffer are incubated together with AR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AR concentration since AR from samples and AR-HRP conjugate compete for the anti-AR antibody binding site. Since the number of sites is limited, as more sites are occupied by AR from the sample, fewer sites are left to bind AR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AR concentration in each sample is interpolated from this standard curve.

NCBI Description

The androgen receptor gene is more than 90 kb long and codes for a protein that has 3 major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. The protein functions as a steroid-hormone activated transcription factor. Upon binding the hormone ligand, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and then stimulates transcription of androgen responsive genes. This gene contains 2 polymorphic trinucleotide repeat segments that encode polyglutamine and polyglycine tracts in the N-terminal transactivation domain of its protein. Expansion of the polyglutamine tract causes spinal bulbar muscular atrophy (Kennedy disease). Mutations in this gene are also associated with complete androgen insensitivity (CAIS). Two alternatively spliced variants encoding distinct isoforms have been described. [provided by RefSeq, Jul 2008]
