Intended Uses: This AQP-1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep AQP-1. This ELISA kit for research use only!

Principle of the Assay: AQP-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AQP-1 antibody and an AQP-1-HRP conjugate. The assay sample and buffer are incubated together with AQP-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AQP-1 concentration since AQP-1 from samples and AQP-1-HRP conjugate compete for the anti-AQP-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by AQP-1 from the sample, fewer sites are left to bind AQP-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AQP-1 concentration in each sample is interpolated from this standard curve.

NCBI Description

integral membrane protein that is a major water transport molecule in the kidney proximal tubule and red blood cells [RGD, Feb 2006]

Uniprot Description

AQP1: Forms a water-specific channel that provides the plasma membranes of red cells and kidney proximal tubules with high permeability to water, thereby permitting water to move in the direction of an osmotic gradient. Belongs to the MIP/aquaporin (TC 1.A.8) family.

Product Notes

The Sheep AQP-1 aqp1 (Catalog #AAA737508) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA737508 ELISA Kit recognizes Sheep AQP-1. It is sometimes possible for the material contained within the vial of "Aquaporin 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
