Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for AGRN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any AGRN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AGRN is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AGRN bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

AGRN: Plays a central role in the formation and the maintenance of the neuromuscular junction (NMJ), the synapse between motor neuron and skeletal muscle. Ligand of the MUSK signaling complex that directly binds LRP4 in this complex and induces the phosphorylation of MUSK, the kinase of the complex. The activation of MUSK in myotubes induces the formation of NMJ by regulating different processes including the transcription of specific genes and the clustering of AChR in the postsynaptic membrane. Defects in AGRN are a cause of myasthenia, limb-girdle, familial (LGM). A congenital myasthenic syndrome characterized by a typical 'limb girdle' pattern of muscle weakness with small, simplified neuromuscular junctions but normal acetylcholine receptor and acetylcholinesterase function.
