Related Product Information for ADAM10 elisa kit
Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ADAM10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ADAM10 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADAM10 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADAM10 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

Members of the ADAM family are cell surface proteins with a unique structure possessing both potential adhesion and protease domains. This gene encodes and ADAM family member that cleaves many proteins including TNF-alpha and E-cadherin. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Cleaves the membrane-bound precursor of TNF-alpha at '76-Ala-|-Val-77' to its mature soluble form. Responsible for the proteolytical release of soluble JAM3 from endothelial cells surface. Responsible for the proteolytic release of several other cell-surface proteins, including heparin-binding epidermal growth-like factor, ephrin-A2 and for constitutive and regulated alpha-secretase cleavage of amyloid precursor protein (APP). Contributes to the normal cleavage of the cellular prion protein. Involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicles, suggesting a vesicle-based protease activity. Controls also the proteolytic processing of Notch and mediates lateral inhibition during neurogenesis. Responsible for the FasL ectodomain shedding and for the generation of the remnant ADAM10-processed FasL (FasL APL) transmembrane form. Also cleaves the ectodomain of the integral membrane proteins CORIN and ITM2B. May regulate the EFNA5-EPHA3 signaling.8 PublicationsManual assertion based on experiment in:Ref.4

Catalytic activity: Endopeptidase of broad specificity.

Cofactor: Binds 1 zinc ion

Product Notes

The Human ADAM10 adam10 (Catalog #AAA704324) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA704324 ELISA Kit recognizes Human ADAM10. It is sometimes possible for the material contained within the vial of "ADAM metallopeptidase domain 10, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
