Biological Activity
Murine Interleukin-1 beta is fully biologically active when compared to standard. The ED50 as determined by the dose-dependent stimulation of murine D10S cells < 0.002ng/ml, corresponding to a Specific Activity of 5 x10e9 IU/mg.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile dH2O, 0.1% HSA or BSA, aliquot and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.

A soluble factor produced by monocytes, macrophages, and other cells which activates T-lymphocytes and potentiates their response to mitogens or antigens. Murine macrophages display a transition from IL-1 beta to IL-1 alpha production during maturation of monocytes into inflammatory macrophages. The synthesis of IL-1 can be induced by other cytokines including TNF-alpha, IFN-alpha, IFN-beta and IFN-g and also by bacterial endotoxins, viruses, mitogens, and antigens. In human skin fibroblasts IL-1 alpha and TNF-alpha induce the synthesis of IL-1beta. In pheochromocytoma cells, NGF induces the synthesis of IL-1.

Function: Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.

Subcellular location: Secreted. Note: The lack of a specific hydrophobic segment in the precursor sequence suggests that IL-1 is released by damaged cells or is secreted by a mechanism differing from that used for other secretory proteins.
