Principle of the Assay: Human Apoptosis Inducing Factor (AIF) ELISA Kit employs a two-site sandwich ELISA to quantitate AIF in samples. An antibody specific for AIF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any AIF present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human AIF detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of AIF bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background/Introduction: Apoptosis-inducing factor (AIF) is a flavin adenine dinucleotide-containing, NADH-dependent oxidoreductase residing in the mitochondrial intermembrane space, which contributes to cell death programmes and participates in the assembly of the respiratory chain. Besides playing a key role in execution of caspase-independent cell death, AIF has emerged as a protein critical for cell survival. Importantly, AIF deficiency leads to severe mitochondrial dysfunction, causing muscle atrophy and neurodegeneration in model organisms as well as in humans.

Uniprot Description

PDCD8: Probable oxidoreductase that has a dual role in controlling cellular life and death; during apoptosis, it is translocated from the mitochondria to the nucleus to function as a proapoptotic factor in a caspase-independent pathway, while in normal mitochondria, it functions as an antiapoptotic factor via its oxidoreductase activity. The soluble form (AIFsol) found in the nucleus induces 'parthanatos' i.e., caspase-independent fragmentation of chromosomal DNA. Interacts with EIF3G,and thereby inhibits the EIF3 machinery and protein synthesis, and activates casapse-7 to amplify apoptosis. Plays a critical role in caspase- independent, pyknotic cell death in hydrogen peroxide-exposed cells. Binds to DNA in a sequence-independent manner. Interacts with XIAP/BIRC4. Interacts (via N-terminus) with EIF3G (via C-terminus). Belongs to the FAD-dependent oxidoreductase family. 4 isoforms of the human protein are produced by alternative splicing.
