Principle of the Assay: The Human Nectin 4 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Nectin 4 in Cell Culture Supernates, Serum, Plasma, Cell Lysates, Tissue Homogenates, Urine. This assay employs an antibody specific for Human Nectin 4 coated on a 96-well plate. Standards and samples are pipetted into the wells and Nectin 4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Nectin 4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Nectin 4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Nectin-4, also known as PVRL4, is an adhesion receptor of the immunoglobulin superfamily and functions in cell-cell adhesion. This gene is mapped to chromosome 1q23.3. The encoded protein contains two immunoglobulin-like (Ig-like) C2-type domains and one Ig-like V-type domain. It is involved in cell adhesion through trans-homophilic and -heterophilic interactions. It is a single-pass type I membrane protein. The soluble form is produced by proteolytic cleavage at the cell surface by the metalloproteinase ADAM17/TACE. The secreted form is found in both breast tumor cell lines and breast tumor patients. Mutations in this gene are the cause of ectodermal dysplasia-syndactyly syndrome type 1, an autosomal recessive disorder. Alternatively spliced transcript variants have been found but the full-length nature of the variant has not been determined.
