Principle of the Assay||The Human IL-23 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-23 in Cell Culture Supernatants, Serum, Plasma, Tissue Homogenates. This assay employs an antibody specific for Human IL-23 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-23 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-23 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-23 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Interleukin-23 subunit alpha (IL-23) is a protein that in humans is encoded by the IL23A gene. IL-23 is a heterodimeric cytokine consisting of two subunits, one called p40, which is shared with another cytokine, IL-12, and another called p19 (the IL-23 alpha subunit). IL-23 is an important part of the inflammatory response against infection. It promotes upregulation of the matrix metalloprotease MMP9, increases angiogenesis and reduces CD8+ T-cell infiltration. Recently, IL-23 has been implicated in the development of cancerous tumors. he International Radiation Hybrid Mapping Consortium mapped the p19 gene to chromosome 12 (stSG47812).

NCBI Description

This gene encodes a subunit of the heterodimeric cytokine interleukin 23 (IL23). IL23 is composed of this protein and the p40 subunit of interleukin 12 (IL12B). The receptor of IL23 is formed by the beta 1 subunit of IL12 (IL12RB1) and an IL23 specific subunit, IL23R. Both IL23 and IL12 can activate the transcription activator STAT4, and stimulate the production of interferon-gamma (IFNG). In contrast to IL12, which acts mainly on naive CD4(+) T cells, IL23 preferentially acts on memory CD4(+) T cells. [provided by RefSeq, Jul 2008]

Uniprot Description

IL23A: Associates with IL12B to form the IL-23 interleukin, a heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to a heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak- Stat signaling cascade, stimulates memory rather than naive T- cells and promotes production of proinflammatory cytokines. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis. Belongs to the IL-6 superfamily.

Product Notes

The Human IL23A il23a (Catalog #AAA8244559) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8244559 ELISA Kit recognizes Human IL23A. AAA Biotech's IL-23 can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (EIA). Researchers should empirically determine the suitability of the IL23A il23a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IL-23, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
