Principle of the Assay: IL-23ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IL-23 antibody and an IL-23-HRP conjugate. The assay sample and buffer are incubated together with IL-23-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-23concentration since IL-23from samples and IL-23-HRP conjugate compete for the anti-IL-23antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-23from the sample, fewer sites are left to bind IL-23-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-23concentration in each sample is interpolated from this standard curve.

This gene encodes a subunit of the heterodimeric cytokine interleukin 23 (IL23). IL23 is composed of this protein and the p40 subunit of interleukin 12 (IL12B). The receptor of IL23 is formed by the beta 1 subunit of IL12 (IL12RB1) and an IL23 specific subunit, IL23R. Both IL23 and IL12 can activate the transcription activator STAT4, and stimulate the production of interferon-gamma (IFNG).
