Principle of the Assay: MAN2A1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MAN2A1 antibody and an MAN2A1-HRP conjugate. The assay sample and buffer are incubated together with MAN2A1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MAN2A1 concentration since MAN2A1 from samples and MAN2A1-HRP conjugate compete for the anti-MAN2A1 antibody binding site. Since the number of sites is limited, as more sites are occupied by MAN2A1 from the sample, fewer sites are left to bind MAN2A1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MAN2A1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is one of two neighboring gene family members that encode mitochondrial enzymes which catalyze the oxidative deamination of amines, such as dopamine, norepinephrine, and serotonin. Mutation of this gene results in Brunner syndrome. This gene has also been associated with a variety of other psychiatric disorders, including antisocial behavior. MAOA: Catalyzes the oxidative deamination of biogenic and xenobiotic amines and has important functions in the metabolism of neuroactive and vasoactive amines in the central nervous system and peripheral tissues. MAOA preferentially oxidizes biogenic amines such as 5-hydroxytryptamine (5-HT), norepinephrine and epinephrine. Defects in MAOA are the cause of Brunner syndrome (BRUNS). Brunner syndrome is a form of X-linked non- dysmorphic mild mental retardation. Male patients are affected by a syndrome of borderline mental retardation and exhibit abnormal behavior, including disturbed regulation of impulsive aggression. Obligate female carriers have normal intelligence and behavior. Belongs to the flavin monoamine oxidase family.
