Principle of the Assay: NUP214 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NUP214 antibody and an NUP214-HRP conjugate. The assay sample and buffer are incubated together with NUP214-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NUP214 concentration since NUP214 from samples and NUP214-HRP conjugate compete for the anti-NUP214 antibody binding site. Since the number of sites is limited, as more sites are occupied by NUP214 from the sample, fewer sites are left to bind NUP214-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NUP214 concentration in each sample is interpolated from this standard curve.

The nuclear pore complex is a massive structure that extends across the nuclear envelope, forming a gateway that regulates the flow of macromolecules between the nucleus and the cytoplasm. Nucleoporins are the main components of the nuclear pore complex in eukaryotic cells. This gene is a member of the FG-repeat-containing nucleoporins. The protein encoded by this gene is localized to the cytoplasmic face of the nuclear pore complex where it is required for proper cell cycle progression and nucleocytoplasmic transport. The 3' portion of this gene forms a fusion gene with the DEK gene on chromosome 6 in a t(6,9) translocation associated with acute myeloid leukemia and myelodysplastic syndrome.
