Principle of the assay: NUP214 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NUP214 antibody and an NUP214-HRP conjugate. The assay sample and buffer are incubated together with NUP214-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NUP214 concentration since NUP214 from samples and NUP214-HRP conjugate compete for the anti-NUP214 antibody binding site. Since the number of sites is limited, as more sites are occupied by NUP214 from the sample, fewer sites are left to bind NUP214-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NUP214 concentration in each sample is interpolated from this standard curve.

NCBI Description

The nuclear pore complex is a massive structure that extends across the nuclear envelope, forming a gateway that regulates the flow of macromolecules between the nucleus and the cytoplasm. Nucleoporins are the main components of the nuclear pore complex in eukaryotic cells. This gene is a member of the FG-repeat-containing nucleoporins. The protein encoded by this gene is localized to the cytoplasmic face of the nuclear pore complex where it is required for proper cell cycle progression and nucleocytoplasmic transport. The 3' portion of this gene forms a fusion gene with the DEK gene on chromosome 6 in a t(6,9) translocation associated with acute myeloid leukemia and myelodysplastic syndrome. Uniprot Description

NUP214: May serve as a docking site in the receptor-mediated import of substrates across the nuclear pore complex. A chromosomal aberration involving NUP214 is found in a subset of acute myeloid leukemia (AML); also known as acute non-lymphocytic leukemia. Translocation t(6;9)(p23;q34) with DEK. It results in the formation of a DEK-CAN fusion gene. A chromosomal aberration involving NUP214 is found in some cases of acute undifferentiated leukemia (AUL). Translocation t(6;9)(q21;q34.1) with SET. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Rat NUP214 nup214 (Catalog #AAA730533) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA730533 ELISA Kit recognizes Rat NUP214. It is sometimes possible for the material contained within the vial of "Nucleoporin 214kDa, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
