Specificity
This assay has high sensitivity and excellent specificity for detection of RAB10. No significant cross-reactivity or interference between RAB10 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between RAB10 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This RAB10 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of MouseRAB10. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: RAB10 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-RAB10 antibody and an RAB10-HRP conjugate. The assay sample and buffer are incubated together with RAB10-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RAB10 concentration since RAB10 from samples and RAB10-HRP conjugate compete for the anti-RAB10 antibody binding site. Since the number of sites is limited, as more sites are occupied by RAB10 from the sample, fewer sites are left to bind RAB10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RAB10 concentration in each sample is interpolated from this standard curve.

NCBI Description

RAB10 belongs to the RAS (see HRAS; MIM 190020) superfamily of small GTPases. RAB proteins localize to exocytic and endocytic compartments and regulate intracellular vesicle trafficking (Bao et al., 1998 [PubMed 9918381]).[supplied by OMIM, Mar 2009]

Uniprot Description

RAB10: a protein of the GTPase superfamily, Rab family. May be involved in vesicular trafficking and neurotransmitter release. Localized to the inner face of the cell membrane by a lipid-anchor.

Protein type: G protein, monomeric; G protein; G protein, monomeric, Rab

Chromosomal Location of Human Ortholog: 2p23.3

Cellular Component: Golgi apparatus; endoplasmic reticulum membrane; recycling endosome; phagocytic vesicle membrane; focal adhesion; cytoplasmic vesicle membrane; recycling endosome membrane; exocyst; plasma membrane; endosome membrane; trans-Golgi network; endosome

Product Notes

The Mouse RAB10 rab10 (Catalog #AAA7204709) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7204709 ELISA Kit recognizes Mouse RAB10. It is sometimes possible for the material contained within the vial of "Ras-related protein Rab-10 (RAB10), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
