Related Product Information for Tyms recombinant protein
Thymidylate synthase is an intracellular enzyme critical for de novo synthesis of DNA. This function maintains the dTMP(thymidine-5-prime monophosphate) pool critical for DNA replication and repair. In cancer, expression of this protein is often elevated and becomes further elevated as a result of treatment with the most commonly used chemotherapeutic, 5-fluorouracil (5-FU). Resistance or lack of response to 5-FU is attributed to the elevation of thymidylate synthase activity. Recombinant human Thymidylate synthase protein, fused to His-tag at N-terminus, was expressed in E. coli and purified by using conventional chromatography techniques.

NCBI Description

This gene encodes an enzyme that catalyzes the methylation of deoxyuridylate to deoxythymidylate using 5,10-methylenetetrahydrofolate as a cofactor. This function maintains the thymidine-5-prime monophosphate concentration critical for DNA replication and repair. The encoded enzyme is a target for cancer chemotherapeutic agents. The majority of transcripts for this gene lack a 3' UTR (PMID: 3022294, 3444407). The stop codon in these transcripts is UAA, compared to the UAG found in the genome and longer transcripts, as the polyA site is located within the stop codon (PMID: 3444407, 2157203). A related pseudogene has been identified on chromosome 10. [provided by RefSeq, Mar 2010]

Uniprot Description

TYMS: an enzyme in the thymidylate synthase family. Polymorphisms serve as prognostic markers for determining modality of treatment in certain cancers. Genotyping helps predict toxicity to 5-FU-based chemotherapy in patients with colorectal cancer and the response to platinum-based chemotherapy in advanced NSCLC. A common polymorphism is a determinant of red blood cell folate and homocysteine concentrations. Homodimer.

Molecular Function: mRNA binding; transferase activity; methyltransferase activity; protein homodimerization activity; nucleotide binding; drug binding; cofactor binding; thymidylate synthase activity; folic acid binding

It is sometimes possible for the material contained within the vial of "Thymidylate synthase, aa1-313 Human, His tag, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
