Application Notes
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect mouse PILR-L. The detection limit for rmPILR-L is approximately 2 ng/lane under non-reducing and reducing conditions, respectively., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse PILR-L. The detection limit for rmPILR-L is approximately 0.1 ng/well. In this format, this antibody shows less than 1% crossreactivity with rmPILR- and rmPILR-., Optimal dilutions should be determined by each laboratory for each application.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

PILR-L (also known as CD99 and MIC2) is a 125 amino acid, 24-30 kD glycosylated transmembrane glycoprotein expressed as a homodimer on the surface of endothelial, T and B cells, monocytes and stem cells. PILR-L has been implicated in processes like homotypic cell adhesion, diapedesis and oncosupression. The extracellular domain of the mouse PILR-L protein used as immunogen is 45% identical to the human counterpart.

Plays a role in a late step of leukocyte extravasation helping leukocytes to overcome the endothelial basement membrane. Acts at the same site as, but independently of, PECAM1. Involved in T-cell adhesion processes. Belongs to the CD99 family. 2 isoforms of the human protein are produced by alternative splicing.
