Application Notes
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect human Tenascin R. The detection limit for rhTenascin R is approximately 2 ng/lane and 20 ng/lane under non-reducing and reducing conditions, respectively., Immunohistochemistry: This antibody will detect human Tenascin R in cells and tissues. The working dilution is 2-5ug/ml., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human Tenascin R. The detection limit for rhTenascin R is approximately 0.3 ng/well., Optimal dilutions should be determined by each laboratory for each application.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Tenascin R is a 180 kD, 1327 amino acid, extracellular matrix glycoprotein belonging to the Tenascin family of adhesion proteins. Tenascin R is selectively expressed in the central nervous system, affecting neurite outgrowth and synaptic functions. Tenascin R isoform 2 shows a deletion of fibronectin type III repeat #6 (aa # 773-862). The human Tenascin R isoform 1 amino acid sequence is 94% identical to the corresponding canine and mouse sequences.

Uniprot Description

Function: Neural extracellular matrix (ECM) protein involved in interactions with different cells and matrix components. Involved in cell attachment and neurite formation. Interaction with CNTN1 enhances the neurite outgrowth. Ref.2

Subunit structure: Forms homodimers and homotrimers. Interacts with CNTN1, NFASC and CSPG5. Ref.1 Ref.2 Ref.3 Ref.4 Ref.5

Subcellular location: Secreted › extracellular space › extracellular matrix. Note: May be attach to the cell surface of neural cells.

Tissue specificity: Brain specific. Ref.1

Developmental stage: Expression weakly detectable at E6 embryo, reaches a maximum at E16 and declines in the adult. Ref.1

Domain: The N-terminus cysteine-rich segment may mediate the formation of oligomers. The fibronectin type-III 2-3 mediate the binding to contactin 1. The fibronectin type-III 9 mediates the cell attachment. The fibronectin type-III 2-5 mediate NFASC binding. The fibrinogen C-terminal domain mediates interaction with CSPG5. Ref.5
