Application Notes
Suitable for use in Western Blot, Immunoprecipitation and Immunocytyochemistry.
Dilution: Western Blot Analysis: 1:2000-1:10,000 detects methylated Histone H3 in acid extracted proteins from HeLa cells. 1:2000-1:5000 dilution of a previous lot detected methylated Histone H3 in acid extracted proteins from HeLa and tetrahymena cells. HeLa cell acid precipitate was resolved by electrophoresis, transferred to nitrocellulose and probed with a 1:5000 dilution of H5110-14J. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Chromatin Immunoprecipitation: 5ul immunoprecipitates methylated Histone H3 from1x10e6 3T3/A31 cells.
Immunocytochemistry: 1:500 shows positive immunostaining for methylated Histone H3 in L6 cells xed with 4% paraformaldehyde and permeabilized using 0.1% Triton X-100.
Optimcal dilutions should be determined by the researcher.

May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
