Application Notes
Suitable for use in Immunohistochemistry, Western Blot, and ELISA.
Dilution: Immunohistochemistry: 10ug/ml detects Wnt-8b on mouse fetal brain tissue sections. Sections were fixed with PBS containing 4% paraformaldehyde for 20 minutes at RT and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA for 45 minutes at RT. After blocking, cells were incubated with diluted W3037-10 overnight at 4 degree C followed by Rhodamine Red-coupled anti-rat IgG at RT in the dark for one hour. Between each step, cells were washed with PBS containing 0.1% BSA.
Western Blot: 1-2ug/ml with the appropriate secondary reagents detects human or mouse Wnt-8b. Using a colorimetric detection system, the detection limit is ~25ng/lane under non-reducing and reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold.
Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~5ng/well.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute by adding sterile 40-50% glycerol, PBS. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 95%, 86% and 71% amino acid identity to the mouse, zebrafish and Xenopus Wnt8B proteins, respectively. The expression patterns of the human and mouse genes appear identical and are restricted to the developing brain. The chromosomal location of this gene to 10q24 suggests it as a candidate gene for partial epilepsy. [provided by RefSeq]
