In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against human plasma a single precipitin line is obtained and shows a reaction of full identity with the purified immunogen. The antiserum does not react with any other protein component of human serum or plasma.

Immunogen
Cortisol binding globulin is an alpha-1 glycoprotein with a molecular weight of 55,700. It is bonding and transporting cortisol in plasma. The mean concentration in adult serum is about 50 ug/ml. Elevated levels are observed in pregnancy. Freund's complete adjuvant is used in the first step of the immunization procedure.
Adsorption
Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies reacting with other human serum proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.

Antibody titre
Precipitin titre not less than 1:16 when tested against normal human serum in agar-block immunodiffusion titration.
Intended use
In precipitating techniques as immunoelectrophoresis and single and double radial immunodiffusion (Mancini, Ouchterlony) to identify the presence of cortisol binding globulin or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibody-binding techniques as ELISA and Western blotting if proper controls are included.
Directions for use
In immunoelectrophoresis use 2 III human plasma or equivalent against 120 III antiserum. In double radial immunodiffusion use a rosette arrangement with 10 III antiserum in 3 mm diameter center well and 2 III plasma samples (neat and serially diluted) in 2 mm diameter peripheral wells. In single radial immunodiffusion use 1 percent antiserum in the gel.

Vial with 1 mllyophilized antiserum.
Caution
This antiserum should be handled by qualified persons only and appropriate precautions should be taken in its handling and disposal, and of all associated materials. For in vitro research purposes only.
Preparation and Storage
Lyophilized at +4 degree C = at least 10 years
Reconstituted at or below -20 degree C = 3-5 years
Reconstituted at +4 degree C = 7 days

Handling:
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4 degree C; prolonged storage at or below -20 degree C. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum.
Diluted antiserum should be stored at +4 degree C, not refrozen, and preferably used the same day.

This gene encodes an alpha-globulin protein with corticosteroid-binding properties. This is the major transport protein for glucorticoids and progestins in the blood of most vertebrates. The gene localizes to a chromosomal region containing several closely related serine protease inhibitors which may have evolved by duplication events. [provided by RefSeq, Jul 2008]

Defects in SERPINA6 are a cause of corticosteroid-binding globulin deficiency (CBG deficiency). CBG deficiency is an extremely rare hereditary disorder characterized by reduced corticosteroid-binding capacity with normal or low plasma corticosteroid-binding globulin concentration, and normal or low basal cortisol levels associated with hypo/hypertension and muscle fatigue. Belongs to the serpin family.
