Scientific Background: STAT4 (signal transducer and activator of transcription 4) is a member of the STAT family that is phosphorylated by receptor-associated protein kinases. STATs form homo- or heterodimers and translocate to the cell nucleus where they act as transcription activators. T helper (Th) cells differentiation is regulated by STAT4 (1). STAT4 controls T1/T2 balance during the response to live influenza virus and isolated viral proteins. The differentiation of gamma interferon (IFN-gamma)-producing Th1 and Tc1 cells after inoculation of live virus occurred independently of STAT4 expression. STAT4 has been shown to control pathogenesis of chronic obstructive pulmonary disease (2).

NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is essential for mediating responses to IL12 in lymphocytes, and regulating the differentiation of T helper cells. Mutations in this gene may be associated with systemic lupus erythematosus and rheumatoid arthritis. Alternate splicing results in multiple transcript variants that encode the same protein. Uniprot Description
Function: Carries out a dual function: signal transduction and activation of transcription. Involved in IL12 signaling.
Subunit structure: Forms a homodimer or a heterodimer with a related family member. Interacts with ARL2BP
By similarity. The SH2 domain interacts, in vitro, with IL12RB2 via a short cytoplasmic domain. Ref.4 Ref.5
Subcellular location: Cytoplasm. Nucleus. Note: Translocated into the nucleus in response to phosphorylation.
Post-translational modification: Tyrosine phosphorylated. Serine phosphorylation is also required for maximal transcriptional activity
By similarity.
Involvement in disease: Systemic lupus erythematosus 11 (SLEB11) [MIM:612253]: A chronic, relapsing, inflammatory, and often febrile multisystemic disorder of connective tissue, characterized principally by involvement of the skin, joints, kidneys and serosal membranes. It is of unknown etiology, but is thought to represent a failure of the regulatory mechanisms of the autoimmune system. The disease is marked by a wide range of system dysfunctions, an elevated erythrocyte sedimentation rate, and the formation of LE cells in the blood or bone marrow.Note: Disease susceptibility is associated with variations affecting the gene represented in this entry. Ref.6 Ref.8Rheumatoid arthritis (RA) [MIM:180300]: An inflammatory disease with autoimmune features and a complex genetic component. It primarily affects the joints and is characterized by inflammatory changes in the synovial membranes and articular structures, widespread fibrinoid degeneration of the collagen fibers in mesenchymal tissues, and by atrophy and rarefaction of bony structures.Note: Disease susceptibility is associated with variations affecting the gene represented in this entry. Ref.8
Sequence similarities: Belongs to the transcription factor STAT family.Contains 1 SH2 domain.

Product Notes
The STAT4 stat4 (Catalog #AAA515122) is a Recombinant Protein produced from Sf9 insect cells and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is His tag!!Expression System||Sf9 insect cells using baculovirus!!Source Note||Recombinant full-length human STAT4 was expressed by baculovirus in Sf9 insect cells. AAA Biotech's STAT4 can be used in a range of immunoassay formats including, but not limited to, Kinase Assay, Western Blot (WB). Researchers should empirically determine the suitability of the STAT4 stat4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "STAT4, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
