Related Product Information for anti-STAT4 (pY693) antibody
STAT proteins (Signal transducer and activator of transcription) belong to a family of cytoplasmic transcription factors that can be phosphorylated by a lignad binding to its cell surface receptor. Activation of STAT4 induces dimerization, nuclear translocation and DNA binding. STAT4 protein expression is restricted to the thymus, spleen and testis. Both tyrosine and serine residues on STAT4 become phosphorylated in response to IL-12 or IFN-alpha although serine phosphorylation is not required for DNA binding. STAT4 is activated in T-cells in response to the cytokine interleukin-12 (IL-12), and in NK cells by IL-2. STAT4-deficient mice display impaired IL-12 development of Th1 cells and enhanced development of Th2 cells.

NCBI Description

The protein encoded by this gene is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is essential for mediating responses to IL12 in lymphocytes, and regulating the differentiation of T helper cells. [provided by RefSeq]

Uniprot Description

Function: Carries out a dual function: signal transduction and activation of transcription. Involved in IL12 signaling.

Subunit structure: Forms a homodimer or a heterodimer with a related family member. Interacts with ARL2BP

By similarity. The SH2 domain interacts, in vitro, with IL12RB2 via a short cytoplasmic domain. Ref.4 Ref.5

Subcellular location: Cytoplasm. Nucleus. Note: Translocated into the nucleus in response to phosphorylation.

Post-translational modification: Tyrosine phosphorylated. Serine phosphorylation is also required for maximal transcriptional activity

Involvement in disease: Genetic variations in STAT4 are associated with susceptibility to systemic lupus erythematosus type 11 (SLEB11) [

MIM:612253]. Systemic lupus erythematosus (SLE) is a chronic autoimmune disease with a complex genetic basis. SLE is an inflammatory, and often febrile multisystemic disorder of connective tissue characterized principally by involvement of the skin, joints, kidneys, and serosal membranes. It is thought to represent a failure of the regulatory mechanisms of the autoimmune system. Ref.7Genetic variations in STAT4 are a cause of susceptibility to rheumatoid arthritis (RA) [

MIM:180300]. It is a systemic inflammatory disease with autoimmune features and a complex genetic component. It primarily affects the joints and is characterized by inflammatory changes in the synovial membranes and articular structures, widespread fibrinoid degeneration of the collagen fibers in mesenchymal tissues, and by atrophy and rarefaction of bony structures. Ref.7

Sequence similarities: Belongs to the transcription factor STAT family.Contains 1 SH2 domain.

The Rabbit anti Phospho- STAT4 (pTyr 693) reacts with Human, Rat, Mouse, Canine and may cross-react with other species as described in the data sheet. AAA Biotech's STAT4 (pY693) can be used in a range of immunoassay formats including, but not limited to, ELISA, Western Blot, Immunohistochemistry. Western Blot: 0.1-1 ug/ml ELISA: 0.01-0.1 ug/ml Immunoprecipitation: 2-5 ug/ml Immunohistochemistry: 1:50 ug/ml. Researchers should empirically determine the suitability of the STAT4 (pY693) stat4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "STAT4 (pY693), Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
