Biological Significance
Neurofilaments are the 10nm or intermediate filament proteins found specifically in neurons, and are composed predominantly of three major proteins called NF-L, NF-M and NF-H (1). NF-M is the neurofilament middle or medium molecular weight polypeptide and runs on SDS-PAGE gels at 145-160 kDa, with some variability across species boundaries. Antibodies to NF-M are useful for identifying neuronal cells and their processes in tissue sections and in tissue culture. NF-M antibodies can also be useful to visualize neurofilament accumulations seen in many neurological diseases, such as Amyotrophic Lateral Sclerosis (Lou Gehrig's disease) and Alzheimer's disease (2).
Preparation and Storage
Store at -20 degree C in undiluted aliquots; stable for at least 1 year. Avoid freeze/thaw cycles.

Function: Neurofilaments usually contain three intermediate filament proteins: L, M, and H which are involved in the maintenance of neuronal caliber.

Post-translational modification: There are a number of repeats of the tripeptide K-S-P, NFM is phosphorylated on a number of the serines in this motif. It is thought that phosphorylation of NFM results in the formation of interfilament cross bridges that are important in the maintenance of axonal caliber. Ref.4Phosphorylation seems to play a major role in the functioning of the larger neurofilament polypeptides (NF-M and NF-H), the levels of phosphorylation being altered developmentally and coincidentally with a change in the neurofilament function.Phosphorylated in the head and rod regions by the PKC kinase PKN1, leading to the inhibition of polymerization
