It is recommended to reconstitute the lyophilized IL-1Ra in sterile 18Momega-cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions containing carrier protein such as BSA, HSA or similar.

Protein quantitation was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.91 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the DNAman computer analysis program of protein sequences.

Preparation and Storage
Lyophilized IL-1Ra although stable at room temperature for 3 weeks, should be stored desiccated below -180 degree C. Upon reconstitution Interleukin-3 should be stored at 40C between 2-7 days and for future use below -180 degree C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

Interleukin-1 receptor antagonist (IL-1Ra) is a naturally occurring inflammatory inhibitor protein. It inhibits the activity of IL-1a and IL-1b by competitively blocking binding to their type I and type II receptors. IL-1Ra is produced by corneal epithelial cells, monocytes, neutrophils, macrophages, and fibroblasts. Therapeutically, IL-1Ra may help in the treatment of sepsis, cachexia, rheumatoid arthritis, chronic myelogenous leukemia, asthma, psoriasis, and inflammatory bowel disease. After binding to interleukin-1 associates with the corecptor IL1RAP to form the high affinity interleukin-1 receptor complex which mediates interleukin-1-dependent activation of NF-kappa-B, MAPK and other pathways. Signaling involves the recruitment of adapter molecules such as TOLLIP, MYD88, and IRAK1 or IRAK2 via the respective TIR domains of the receptor/coreceptor subunits. Binds ligands with comparable affinity and binding of antagonist IL1RN prevents association with IL1RAP to form a signaling complex. Belongs to the interleukin-1 receptor family.
