Intended Uses: This IL-1RA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat IL-1RA. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: IL-1RA ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IL-1RA antibody and an IL-1RA-HRP conjugate. The assay sample and buffer are incubated together with IL-1RA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-1RA concentration since IL-1RA from samples and IL-1RA-HRP conjugate compete for the anti-IL-1RA antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-1RA from the sample, fewer sites are left to bind IL-1RA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-1RA concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a cytokine receptor that belongs to the interleukin 1 receptor family. This protein is a receptor for interleukin alpha (IL1A), interleukin beta (IL1B), and interleukin 1 receptor, type I(IL1R1/IL1RA). It is an important mediator involved in many cytokine induced immune and inflammatory responses. This gene along with interleukin 1 receptor, type II (IL1R2), interleukin 1 receptor-like 2 (IL1RL2), and interleukin 1 receptor-like 1 (IL1RL1) form a cytokine receptor gene cluster in a region mapped to chromosome 2q12. IL-1RA: Receptor for IL1A, IL1B and IL1RN. After binding to interleukin-1 associates with the corecptor IL1RAP to form the high affinity interleukin-1 receptor complex which mediates interleukin-1-dependent activation of NF-kappa-B, MAPK and other pathways. Signaling involves the recruitment of adapter molecules such as TOLLIP, MYD88, and IRAK1 or IRAK2 via the respective TIR domains of the receptor/coreceptor subunits. Binds ligands with comparable affinity and binding of antagonist IL1RN prevents association with IL1RAP to form a signaling complex. Belongs to the interleukin-1 receptor family.
