Immunohistochemical Protocol
(Recommended Immunohistochemical Protocol Pretreatment 1. Cut and mount 3-4 micron formalin-fixed paraffin-embedded tissues on positive charged slides. 2. Air dry for 2 hours at 58° C. 3. Oeparaffinize, dehydrate and rehydrate tissues. 4. Subject tissues to heat epitope retrieval using a suitable retrieval solution such as ImmunoDNA Retriever with Citrate or EDTA. 5. Any of three heating methods may be used: a. Electric Pressure Cooker Place standoff rack at base of pressure cooker. Add 1-2 inches of distilled water to the pressure cooker and turn heat to high, and incubate for 15 minutes. Open and immediately transfer slides toroom temperature. b. Water Bath Method Place tissues/slides in a pre-warmed staining dish or coplin jar containing the Immuno DNA Retriever with Citrate or EDTA in a water bath set at 95°-99° C.lncubate for 30-60 minutes. c. Conventional Steamer Method Place tissues/slides in a pre-warmed staining dish or coplin jar containing the ImmunoDNA Retriever with Citrate or EDTA in a Steamer, cover and steam for 30-60 minutes. 6. After heat treatment, transfer slides in ImmunoDNA Retriever with Citrate or EDTA to room temperature and let stand for 15-20 minutes. 7. Wash slides with IHC wash buffer or 01 water. 8. Continue IHC staining protocol. Immunohistochemical Protocol)
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This nuclear enzyme is involved in processes such as chromosome condensation, chromatid separation, and the relief of torsional stress that occurs during DNA transcription and replication. It catalyzes the transient breaking and rejoining of two strands of duplex DNA which allows the strands to pass through one another, thus altering the topology of DNA. Two forms of this enzyme exist as likely products of a gene duplication event. The gene encoding this form, alpha, is localized to chromosome 17 and the beta gene is localized to chromosome 3. The gene encoding this enzyme functions as the target for several anticancer agents and a variety of mutations in this gene have been associated with the development of drug resistance. Reduced activity of this enzyme may also play a role in ataxia-telangiectasia. [provided by RefSeq, Jul 2010]

Product Notes
The Topoisomerase IIa (Catalog #AAA370150) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Topoisomerase IIa RMab reacts with Human and may cross-react with other species as described in the data sheet. It is sometimes possible for the material contained within the vial of "Topoisomerase IIa, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
