Immunofluorescence (IF)

(Immunofluorescence image of MBS3000263 stained HEK293T cells. The cells were 4% paraformaldehyde fixed (20 min) and then incubated in 10% normal goat serum for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody Lamin B1 pAb # MBS3000263 (1:200) at 5ug/ml overnight at +4 degree C. The secondary antibody (Red) was Goat Anti-Rabbit IgG (H+L) Rhodamine (TRITC) used at a 1/2000 dilution for 1h. DAPI was used to stain the cell nuclei (blue).)

An important part of the nucleus is formed by nuclear lamina. Nuclear lamins form a network of filaments at the nucleoplasmic site of the nuclear membrane. Two main subtypes of nuclear lamins can be distinguished, i.e. A type lamins and B type lamins. The A type lamins comprise a set of three proteins arising from the same gene by alternative splicing, i.e. lamin A, lamin C and lamin Adel10, while the B-type lamins include two proteins arising from two distinct genes, i.e. lamin B1 and lamin B2. The nuclear lamins comprise a unique subclass of the intermediate filament protein family. They share a molecular domain organisation with the other intermediate filament proteins in that they are fibrous molecules that have an aminoterminal globular head, a central rod of alpha helices and a carboxy terminal globular domain. Many biochemical and molecular features of lamins have been studied, but their functions remain still largely undetermined. One of the functions ascribed to the lamina is the maintenance of the structural integrity of the nucleus.

NCBI Description

This gene encodes one of the two B-type lamin proteins and is a component of the nuclear lamina. A duplication of this gene is associated with autosomal dominant adult-onset leukodystrophy (ADLD). Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2015]

Uniprot Description

Lamin B1: nuclear lamins are intermediate filament proteins that constitute the lattice-like matrix at the inner face of the nuclear membrane that underlies the nuclear envelop. The lamins, highly conserved throughout evolution, are encoded by three genes in the human: LMNA, LMNB1, and LMNB2. The A-type lamins (lamin A/C) are developmentally regulated and are generally expressed in differentiated cells. The anchoring of chromatin to the nuclear lamina is involved in the control of gene expression and in DNA replication and repair. During mitosis, the nuclear lamina is reversibly disassembled as the lamin proteins are phosphorylated. B-type lamins are phosphorylated within minutes of engaging the IgM surface receptor of resting splenic B cells. Nuclear lamins are cleaved by caspases during apoptosis.
