Intended Uses: For the quantitative detection of Rat DNA mismatch repair protein Mlh1 (MLH1) concentration in serum, plasma, tissue homogenates and other biological fluids.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate MLH1 in Rat serum, plasma, tissue homogenates. An antibody specific for MLH1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MLH1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MLH1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MLH1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

MLH1: a protein involved in the repair of mismatches in DNA. Binds PMS2 or MLH1 and MLH3. Part of the BRCA1-associated genome surveillance complex (BASC), which contains BRCA1, MSH2, MSH6, MLH1, ATM, BLM, PMS2 and the RAD50- MRE11-NBS1 protein complex. This association could be a dynamic process changing throughout the cell cycle and within subnuclear domains. Interacts with MBD4. Interacts with EXO1. Defects in MLH1 are the cause of hereditary non-polyposis colorectal cancer type 2; Turcot syndrome, an autosomal dominant disorder characterized by malignant tumors of the brain; Muir-Torre syndrome (MTS), a rare autosomal dominant disorder characterized by sebaceous neoplasms and visceral malignancy; lobular carcinoma in situ; endometrial cancer; and non-polyposis colorectal cancer.
