Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate FURIN in samples. An antibody specific for FURIN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyFURIN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FURIN is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FURIN bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a calcium-dependent serine endoprotease that proteolytically activates different proprotein substrates traversing the secretory pathway. The encoded protein undergoes proteolytic autoactivation during which an N-terminal propeptide is cleaved to generate the mature protein. Mice lacking the encoded protein die at an embryonic stage and display hemodynamic insufficiency, cardiac ventral closure defect, axial rotation defect and abnormal yolk sac vasculature. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2015]
