• Test Principle: This assay employs a two-site sandwich ELISA to quantitate MAGEA9 in samples. An antibody specific for MAGEA9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAGEA9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAGEA9 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAGEA9 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a member of the MAGEA gene family. The members of this family encode proteins with 50 to 80% sequence identity to each other. The promoters and first exons of the MAGEA genes show considerable variability, suggesting that the existence of this gene family enables the same function to be expressed under different transcriptional controls.
