Preparation and Storage

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Calcium/Calmodulin Dependent Protein Kinase II Alpha (CAMK2a). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Calcium/Calmodulin Dependent Protein Kinase II Alpha (CAMK2a). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Calcium/Calmodulin Dependent Protein Kinase II Alpha (CAMK2a), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Calcium/Calmodulin Dependent Protein Kinase II Alpha (CAMK2a) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

The product of this gene belongs to the serine/threonine protein kinases family, and to the Ca(2+)/calmodulin-dependent protein kinases subfamily. Calcium signaling is crucial for several aspects of plasticity at glutamatergic synapses. This calcium calmodulin-dependent protein kinase is composed of four different chains: alpha, beta, gamma, and delta. The alpha chain encoded by this gene is required for hippocampal long-term potentiation (LTP) and spatial learning. In addition to its calcium-calmodulin (CaM)-dependent activity, this protein can undergo autophosphorylation, resulting in CaM-independent activity. Two transcript variants encoding distinct isoforms have been identified for this gene. [provided by RefSeq, Nov 2008]

The holoenzyme is composed of four different chains: alpha, beta, gamma, and delta. The different chains assemble into homo- or heteromultimeric holoenzymes composed of 8 to 12 subunits. May interact with BAALC, MPDZ, SYN1 and synGAP. 2 isoforms of the human protein are produced by alternative splicing.
