Specificity
This item recognises human Protein tyrosine kinase-like 7 (PTK7), a member of the receptor tyrosine kinase family, otherwise known as Colon Carcinoma Kinase 4 (CCK4). PTK7 is highly expressed in the kidney, liver, lung, pancreas and melanocytes with weaker expression being detected in the thyroid gland, ovaries, brain, heart and skeletal muscle.

PTK7 may play a role in embryonic development. It is thought to function as a cell adhesion molecule and as a novel regulator of the planar cell polarity (PCP) pathway in vertebrates.

Studies also suggest that PTK7 may be involved in the tumour progression. A loss of PTK7 expression has been observed in melanoma cell lines, whereas increased levels of PTK7 have been detected in high-throughput acute myeloid leukaemia samples. In addition, PTK7 is not expressed in normal colon but has been detected in colon carcinomas and as such is a potential marker for tumour progression.

NCBI Description

This gene encodes a member of the receptor protein tyrosine kinase family of proteins that transduce extracellular signals across the cell membrane. The encoded protein lacks detectable catalytic tyrosine kinase activity, is involved in the Wnt signaling pathway and plays a role in multiple cellular processes including polarity and adhesion. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Jul 2012]

Uniprot Description

CCK4: Inactive tyrosine kinase involved in Wnt signaling pathway. Component of both the non-canonical (also known as the Wnt/planar cell polarity signaling) and the canonical Wnt signaling pathway. Functions in cell adhesion, cell migration, cell polarity, proliferation, actin cytoskeleton reorganization and apoptosis. Has a role in embryogenesis, epithelial tissue organization and angiogenesis. Belongs to the protein kinase superfamily. Tyr protein kinase family. Insulin receptor subfamily. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes

The PTK7 ptk7 (Catalog #AAA222895) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI HUMAN PTK7 (N-TERMINAL) reacts with Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's PTK7 can be used in a range of immunoassay formats including, but not limited to, Immunohistology Paraffin*, Western Blot (WB). Western Blot: This item detects a band of approximately 86kDa in colon tissue lysates. Immunohistology - Paraffin: Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. Western Blotting: Minimum Dilution: 0.5; Maximum Dilution: 1.0ug/ml. Researchers should empirically determine the suitability of the PTK7 ptk7 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PTK7, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
