Antiserum to Wnt-3 was raised by repeated immunisation of goats with highly purified antigen. Purified IgG was prepared by affinity chromatography using the immunizing peptide.

Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Goat anti Wnt-3 antibody recognises both human and mouse proto-oncogene Wnt-3, a member of the Wnt family, and ligand of the frizzled family of seven transmembrane receptors.Wnt-3, otherwise known as Int-4, is involved in cell fate decisions and patterning during embryogenesis, cell signalling during neural tube development, and implicated in the tumorogenesis of several cancers including breast, lung, hepatocellular(HCC), and gastric carcinoma.Over-expression of Wnt-3 playes a pivotal role in the resistance of HER2-overexpressing breast tumors to the breast cancer treatment drug Trastuzumab, also known as Herceptin (Wu et al. 2012). The effects of Wnt-3 over-expression included the transactivated expression of EGFR (epidermal growth factor receptor) an increased expression of B-catenin within the nucleus, an increase in N-cadherin and the neural crest transcription factor SLUG, as well as promoting an EMT (epithelial to mesenchymal transition) response. Goat anti Wnt-3 polyclonal antibody exhibits strong fibrous staining in myoepithelial cells of human breast tissue in paraffin sections. Goat anti Wnt-3 polyclonal antibody has been shown to stain mouse hippocampal primary astrocytes in ICC/IF studies, which investigated the role of Wnt-3 levels and Wnt-3 expressing astrocytes, in the regulation of age-associated decline in adult neurogenesis (Okamoto et al. 2011).

NCBI Description

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 98% amino acid identity to mouse Wnt3 protein, and 84% to human WNT3A protein, another WNT gene product. The mouse studies show the requirement of Wnt3 in primary axis formation in the mouse. Studies of the gene expression suggest that this gene may play a key role in some cases of human breast, rectal, lung, and gastric cancer through activation of the WNT-beta-catenin-TCF signaling pathway. This gene is clustered with WNT15, another family member, in the chromosome 17q21 region. [provided by RefSeq, Jul 2008]

WNT3: Ligand for members of the frizzled family of seven transmembrane receptors. Wnt-3 and Wnt-3a play distinct roles in cell-cell signaling during morphogenesis of the developing neural tube. Defects in WNT3 are the cause of autosomal recessive tetra-amelia (ARTTRA). Tetra-amelia is a rare human genetic disorder characterized by complete absence of all four limbs and other anomalies such as craniofacial, nervous system, pulmonary, skeletal and urogenital defects. Belongs to the Wnt family.
