Flow Cytometry: Use 10ul of the suggested working dilution to label 100ul of human whole peripheral blood.
Immunohistology - Frozen: Minimum Dilution: 1/50; Maximum Dilution: 1/100; Application Note: Overnight incubation with clone 44H6 is recommended for optimal results. The epitope recognised by this antibody is reported to be sensitive to formaldehyde fixation and tissue processing. MyBioSource recommends the use of acetone fixation for frozen sections.

Mouse anti Human CD49d monoclonal antibody, clone 44H6 recognizes human CD49d also known as integrin alpha-4 or VLA-4 subunit alpha. CD49d is a ~150kDa single pass type 1 transmembrane glycoprotein with seven FG-GAP repeats, characteristic of alpha integrins, in its extracellular domain. CD49d can be proteolytically cleaved to yield framents of 80 and 70kDa (Hemler et al. 1987). CD49d associates with either CD29 to form VLA-4 or with Integrin beta-7 to form The Peyer patches-specific homing receptor LPAM-1, involved in the lymphocyte migration and homing to gut-associated lymphoid tissue (Sackstein 2006) through its interaction with MadCam-1, preferentially expressed on Peyer's patch high endothelial venules and postcapillary venules in lamina propria (Briskin et al. 1997) Mouse anti human CD49d, clone 44H6 is reported to induce homotypic aggregation of pre-B cell lines including HOON and NALM-6 (Letarte et al. 1993).

NCBI Description

The product of this gene belongs to the integrin alpha chain family of proteins. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. This gene encodes an alpha 4 chain. Unlike other integrin alpha chains, alpha 4 neither contains an I-domain, nor undergoes disulfide-linked cleavage. Alpha 4 chain associates with either beta 1 chain or beta 7 chain. [provided by RefSeq, Jul 2008]

ITGA4: Integrins alpha-4/beta-1 (VLA-4) and alpha-4/beta-7 are receptors for fibronectin. They recognize one or more domains within the alternatively spliced CS-1 and CS-5 regions of fibronectin. They are also receptors for VCAM1. Integrin alpha- 4/beta-1 recognizes the sequence Q-I-D-S in VCAM1. Integrin alpha- 4/beta-7 is also a receptor for MADCAM1. It recognizes the sequence L-D-T in MADCAM1. On activated endothelial cells integrin VLA-4 triggers homotypic aggregation for most VLA-4-positive leukocyte cell lines. It may also participate in cytolytic T-cell interactions with target cells. Heterodimer of an alpha and a beta subunit. The alpha subunit can sometimes be cleaved into two non-covalently associated fragments. Alpha-4 associates with either beta-1 or beta-7. Alpha-4 interacts with PXN, LPXN, and TGFB1I1/HIC5. Interacts with CSPG4 through CSPG4 chondroitin sulfate glycosaminoglycan. Belongs to the integrin alpha chain family.
