Intended Uses: This sandwich kit is for the accurate quantitative detection of human Cystatin-Ar (also known as CSTA/STF1/STFA) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human CSTA/STF1/STFA antibody. CSTA/STF1/STFA present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human CSTA/STF1/STFA Antibody is added and binds to CSTA/STF1/STFA in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CSTA/STF1/STFA antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human CSTA/STF1/STFA. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins, and kininogens. This gene encodes a stefin that functions as a cysteine protease inhibitor, forming tight complexes with papain and the cathepsins B, H, and L. The protein is one of the precursor proteins of cornified cell envelope in keratinocytes and plays a role in epidermal development and maintenance. Stefins have been proposed as prognostic and diagnostic tools for cancer. [provided by RefSeq, Jul 2008]

This is an intracellular thiol proteinase inhibitor. Has an important role in desmosome-mediated cell-cell adhesion in the lower levels of the epidermis.
