Intended Uses: This sandwich kit is for the accurate quantitative detection of human Neurogranin (also known as NRGN) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human NRGN antibody. NRGN present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human NRGN Antibody is added and binds to NRGN in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated NRGN antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human NRGN. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

Neurogranin (NRGN) is the human homolog of the neuron-specific rat RC3/neurogranin gene. This gene encodes a postsynaptic protein kinase substrate that binds calmodulin in the absence of calcium. The NRGN gene contains four exons and three introns. The exons 1 and 2 encode the protein and exons 3 and 4 contain untranslated sequences. It is suggested that the NRGN is a direct target for thyroid hormone in human brain, and that control of expression of this gene could underlay many of the consequences of hypothyroidism on mental states during development as well as in adult subjects.
