Intended Uses: This sandwich kit is for the accurate quantitative detection of human Claudin 5 (also known as CLDN5) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.
Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human CLDN5 antibody. CLDN5 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human CLDN5 Antibody is added and binds to CLDN5 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CLDN5 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human CLDN5. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

This gene encodes a member of the claudin family. Claudins are integral membrane proteins and components of tight junction strands. Tight junction strands serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets. Mutations in this gene have been found in patients with velocardiofacial syndrome. Alternative splicing results in multiple transcript variants encoding distinct isoforms.
