Related Product Information for VWF elisa kit
Introduction: Willebrand factor (vWF) is a blood glycoprotein involved in hemostasis. It is deficient or defective in von Willebrand disease and is involved in a large number of other diseases, including thrombotic thrombocytopenic purpura, Heyde's syndrome, and possibly hemolytic-uremic syndrome. vWF is produced constitutively in endothelium, megakaryocytes, and subendothelial connective tissue. The basic vWF monomer is a 2050 amino acid protein. Every monomer contains a number of specific domains with a specific function. Monomers are subsequently N-glycosylated, arranged into dimers in the endoplasmic reticulum and into multimers in the Golgi apparatus by crosslinking of cysteine residues via disulfide bonds. With respect to the glycosylation, vWF is one of the few proteins that carry ABO blood group system antigens. Multimers of vWF can be extremely large, >20,000 kDa, and consist of over 80 subunits of 250 kDa each. Only the large multimers are functional. Some cleavage products that result from Vwf production are also secreted but probably serve no function. Von Willebrand factor is not an enzyme and therefore has no catalytic activity. Its primary function is binding to other proteins, particularly Factor VIII and it is important in platelet adhesion to wound sites. vWF binds to a number of cells and molecules. The most important ones are Factor VIII, collagen, platelet gpIb, other platelet receptors et al.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to vWF. Standards or samples are then added to the appropriate microtiter plate wells with a biotinconjugated antibody preparation specific for vWF and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain vWF, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of vWF in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uniprot Description

Function: Important in the maintenance of hemostasis, it promotes adhesion of platelets to the sites of vascular injury by forming a molecular bridge between sub-endothelial collagen matrix and platelet-surface receptor complex GPIb-IX-V. Also acts as a chaperone for coagulation factor VIII, delivering it to the site of injury, stabilizing its heterodimeric structure and protecting it from premature clearance from plasma By similarityManual assertion inferred from sequence similarityUniProtKB:P04275

Product Notes

The Mouse VWF vwf (Catalog #AAA703001) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA703001 ELISA Kit recognizes Mouse VWF. It is sometimes possible for the material contained within the vial of "von Willebrand factor, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
