Scientific Background: UBE2R2 (UBC3B) or ubiquitin-conjugating enzyme E2R 2 encodes a protein similar to the E2 ubiquitin conjugating enzyme UBC3/CDC34. CK2-dependent phosphorylation of this ubiquitin-conjugating enzyme functions by regulating beta-TrCP substrate recognition and induces its interaction with beta-TrCP therby enhancing beta-catenin degradation. CK2-dependent phosphorylation of CDC34 and UBC3B functions by regulating BTRC substrate recognition (1). UBE2R2 complements a yeast cdc34 temperature-sensitive mutant. Deletion and site-directed mutagenesis demonstrated that CK2 phosphorylated UBE2R2 in the C-terminal domain at serine-233; mutagenesis of the corresponding serine residue at position 231 of human CDC34/UBC3 showed that this was the major CK2-phosphorylation site in this protein.

NCBI Description

Protein kinase CK2 is a ubiquitous and pleiotropic Ser/Thr protein kinase involved in cell growth and transformation. This gene encodes a protein similar to the E2 ubiquitin conjugating enzyme UBC3/CDC34. Studies suggest that CK2-dependent phosphorylation of this ubiquitin-conjugating enzyme functions by regulating beta-TrCP substrate recognition and induces its interaction with beta-TrCP, enhancing beta-catenin degradation. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. In vitro catalyzes monoubiquitination and 'Lys-48'-linked polyubiquitination. May be involved in degradation of katenin. Ref.1 Ref.8

Catalytic activity: ATP + ubiquitin + protein lysine = AMP + diphosphate + protein N-ubiquityllysine. Ref.1

Pathway: Protein modification; protein ubiquitination.

Subunit structure: When phosphorylated, interacts with beta-TrCP (BTRC). Ref.1

Sequence similarities: Belongs to the ubiquitin-conjugating enzyme family.

Product Notes

The UBE2R2 (UBC3B) ube2r2 (Catalog #AAA515256) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is His tag!!Expression System||E.coli!!Source Note||Recombinant human UBE2R2 (UBC3B) (2-end) was expressed in E. coli cells. AAA Biotech's UBE2R2 (UBC3B) can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the UBE2R2 (UBC3B) ube2r2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "UBE2R2 (UBC3B), Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
